Liverpool put in an expert performance in the first half but Mohamed Salah's opener was cancelled out by Heung-min Son's strike.
Both sides had their chances after the break but the Reds came out on top as Roberto Firmino headed home to put the champions three points clear at the top of the Premier League table.
Jurgen Klopp made just the sole change to the side that drew with Fulham as Rhys Williams replaced Joel Matip in defence after the Cameroonian suffered a back spasm at the weekend.
Roberto Firmino had an excellent chance from Andy Robertson's set-piece as the Brazilian headed goalwards after being left unmarked, but the effort lacked the power to beat Hugo Lloris between the sticks.
Liverpool enjoyed the majority of the ball as Spurs struggled to get their attacking players involved. However, there were a couple of niggling fouls from the visitors as they attempted to disrupt the Reds' rhythm.
With a little under 20 minutes played, replays showed a duel between Mohamed Salah and Eric Dier inside the box saw the Spurs defender use his arm to intercept the ball, but there was no interest from VAR as play went on.
An excellent Reds move concocted by Jordan Henderson and Robertson saw the latter pull the ball back for Salah, whose first-time effort went straight at Lloris.
Liverpool took a deserved lead midway through the half as the ball fell to Salah at the edge of the box, who this time got a slice of good fortune as the ball ricocheted off Toby Alderweireld and looped into the back of the net.
But Spurs hit back against the run of play as Heung-min Son beat the offside trap and gave Alisson the eyes before rolling the ball inside the near post to give Spurs a surprising leveller.
Firmino then forced Lloris into an excellent low save before Sadio Mane got a shot off from the edge of the box, but the strike went straight at the Frenchman.
Spurs had a glorious chance inside the opening minute of the second half as Steven Bergwijn got the better of Trent Alexander-Arnold, but the winger could only skew his effort past the far post.
The visitors showed greater attacking intent as the half wore on as the Reds struggled to create any clear-cut chances, despite some decent moves put together.
Bergwijn got in behind for the second time before hitting the post with a curling strike. Harry Kane then had the best chance of the game thus far as he nodded the ball into the ground and over from less than six yards from a Spurs corner.
Firmino had a strike from the edge of the box as the Reds looked to assert their dominance once more, but his effort went straight into the arms of Lloris.
An excellent interception and pass by Williams played Salah in on goal but the angle was only getting tighter as he rolled the ball comfortably within the keeper's reach.
A clever turn by Mane saw him go close with a powerful strike that went just over the bar as Liverpool turned the screw, prompting Jose Mourinho to bring Sergio Reguilon on for Bergwijn.
With time running out and the Reds seemingly headed for yet another 1-1 draw, Firmino connected with a Robertson corner to score only his fourth goal in his last 28 games with a fantastic header.
The strike was enough for Klopp's side to seal all three points, with the Reds boss visibly ecstatic with his side's performance at full-time.
